Programmed cell death 1 as prognostic marker and therapeutic target in upper gastrointestinal cancers

چکیده

Gastrointestinal (GIs) cancers are among the most common causes of cancer related death, and hence importance for identification novel prognostic/predictive biomarkers detection patients at an early stage, using these to identify targeted therapies improve efficacy existing chemotherapeutic regimens. Programmed cell death 1 has been reported as a potential target in several malignancies, targeting agents being developed, some already approved by FDA, such as: pembrolizumab, Atezolizumab, Nivolumab. Pembrolizumab that have treatment metastatic non-small lung cancer. Here we provide overview mechanism action PD-1/PD-L1, prognostic value current progress clinical trials PD-1/PD-L1 inhibitors, resistant mechanisms underlie inhibitory effect gastrointestinal cancers.

Antagonizing programmed death-1 and programmed death ligand-1 as a therapeutic approach for gastric cancer

Malignant tumor cells are equipped with mechanisms that can help them escape the surveillance by host immune system. Immune checkpoint molecules can transduce coinhibitory signals to immunocompetent cells and exert immunosuppressive roles in antitumor immunity. Programmed death-1 (PD-1) and programmed death ligand-1 (PD-L1) are the two important checkpoint molecules with great potential in targ...

YB-1: oncoprotein, prognostic marker and therapeutic target?

Hanahan and Weinberg have proposed the 'hallmarks of cancer' to cover the biological changes required for the development and persistence of tumours [Hanahan and Weinberg (2011) Cell 144, 646-674]. We have noted that many of these cancer hallmarks are facilitated by the multifunctional protein YB-1 (Y-box-binding protein 1). In the present review we evaluate the literature and show how YB-1 mod...

Programmed Cell Death-1/Programmed Death-ligand 1 Pathway: A New Target for Sepsis

OBJECTIVE Sepsis remains a leading cause of death in many Intensive Care Units worldwide. Immunosuppression has been a primary focus of sepsis research as a key pathophysiological mechanism. Given the important role of the negative costimulatory molecules programmed cell death-1 (PD-1) and programmed death-ligand 1 (PD-L1) in the occurrence of immunosuppression during sepsis, we reviewed litera...

Prognostic significance of programmed death-1 and programmed death-ligand 1 expression in patients with esophageal squamous cell carcinoma

AIMS To evaluate the expression of programmed death-1 (PD-1) and programmed death-ligand 1 (PD-L1) and their clinical and prognostic significance in primary esophageal squamous cell carcinoma (ESCC). RESULTS The expression rate of PD-1 and PD-L1 in ESCC was 33.5% (117/349) and 41.4% (222/536), respectively. PD-L1 expression differed significantly by tumor location, grade, lymph node metastase...

c-Met is a prognostic marker and potential therapeutic target in clear cell renal cell carcinoma.

BACKGROUND Activation of the c-Met pathway occurs in a range of malignancies, including papillary renal cell carcinoma (RCC). Its activity in clear cell RCC is less clear. We investigated c-Met expression and inhibition in a large cohort of RCC tumors and cell lines. METHODS c-Met protein expression was determined by automated quantitative analysis (AQUA) on a tissue microarray (TMA) construc...
